Volatile compounds of Tripleurospermum decipiens from different sites in Turkey

Conference Paper Planta Medica, Volume 81, Issue S01, 2016, Pages S1-S381

Abstract

The genus Tripleurospermum Schultz Bip. is represented in Turkey by 31 taxa, 15 being endemic in Turkey[1-3]. Hydrodistilled essential oils of the dried flowers of Tripleurospermum decipiens (Fisch. & Mey.) Bornm. [Asteraceae] were analyzed by GC-FID and GC-MS. The oils were characterized by the occurrence of matricaria esters as main constituents. Flowers of T. decipiens were collected from two localities in Turkey; A: Adana: Saimbeyli-Yesilkent road, 12.06.2014–TDA; B: Eskişehir: Between Bozüyük- Eskişehir, 26.06.2009–TDB. The essential oils of TDA and TDB were characterized by the occurrence of (2Z,8Z)-matricaria ester (TDA: 57.9%; TDB: 70.0%), ß-sesquiphellandrene (TDA: 10.4%; TDB: 2.7%), (2E,8Z)-matricaria ester (TDA: 8.1%; TDB: 1.7%), (Z)-ß-farnesene (TDA: 7.5%, TDB: 0.3%), (2E,8E)-matricaria ester (TDA: 2.3%, TDB: 0.4%) and (2Z,8E)-matricaria ester (TDA: 1.4%; TDB: 0.5%) as main constituents. Matricaria esters were previously found in other Tripleurospermum species such as T. disciforme (C.A.Mey) Schultz-Bip. [5] and Matricaria perforata Merat (=Tripleurospermum inodorum (L.) Sch. Bip) [6]. To the best of our knowledge, this is the first report on the essential oil composition of Tripleurospermum decipiens.

Unexpected Irregular Monoterpene "Yomogi Alcohol" in the Volatiles of the Lathyrus L. species (Leguminosae) of Cyprus.

Original Article J. Oleo Sci. , Volume 65, Issue 3, 2016, Pages 241-249

Abstract

Lathyrus species including L. ochrus and L. sativus are known for their food, feed and horticultural uses. Despite their widespread uses and cultivation, there is limited information on their chemistry. Previously, only the essential oil composition of L. rotundifolius, L. vernus and volatiles of L. odoratus have been reported. In the present research, volatiles of seven Lathyrus L. species, namely, L. aphaca, L. ochrus, L. cicera, L. sativus, L. gorgonei, L. saxatilis and L. blepharicarpos var. cyprius were analyzed by SPME GC-MS for the first time. Plant materials were collected from five different locations in Cyprus (February-March 2012). The main components of L. aphaca volatiles from four locations were yomogi alcohol 26.1-16.5%, camphor 21.6-10.1%, tetradecane 14.3-0%; L. cicera from five locations were yomogi alcohol 20.3-3.0%, camphor 18.7-2.0%; L. gorgonei from two locations were yomogi alcohol 24.5-13.1%, camphor 17.1-9.0% and L. sativus was yomogi alcohol 11.4%, camphor 9.0%. Yomogi alcohol was not present as the major compound in L. ochrus (2-methyl butanoic acid 7.2%), L. saxatilis (hexanal 7.7%) and L. blepharicarpos var. cyprius ((Z)-3-hexenal 8.6%) volatiles. The volatiles of the Lathyrus species were also compared with each other quantitative and qualitatively using AHC analysis to find out differences among the species. The irregular monoterpene yomogi alcohol is reported from the Lathyrus and the Leguminosae family for the first time. The existence of yomogi alcohol in Lathyrus volatiles points out that the irregular monoterpenes are not restricted solely to Asteraceae family.

11-Hydroxy-2,4-cycloeudesmane from leaf oil of Juglans regia and evaluation of its larvicidal activity

Original Article Natural Product Communications, Volume 11, Issue 10, 2016, Pages 1421-1424

Abstract

Leaves of Juglans regia L. collected from two different locations [Adana (A) and Ankara (B)] from Turkey were subjected to hydrodistillation in a Clevenger type apparatus to yield the essential oils (EOs). GC/MS and GC-FID analyses revealed that the A EO was rich in thymol (23.1%), while caryophyllene oxide(33.8%) was found as the main constituent of B EO. Both contained ?-eudesmol (1.4% - 9.5 %), (E)-geranyl acetone (3.7% - 5.8%) and the eudesmane type constituent juglaterpene A (3.1% - 11.0%). Using a HP Innowax preparative capillary column connected to a preparative fraction collector, an unknown constituent, juglaterpene A (1, 11-hydroxy-2,4-cycloeudesmane), was isolated in a rapid one-step manner with > 94.0% purity. Structure determination was accomplished from 1D- and 2D-NMR spectroscopic data. Oil B demonstrated significant larvicidal activity against 1st instar Aedes aegypti L.

Chemical Composition and Biological Activity of Centaurea baseri: New species from Turkey

Original Article Chemistry and Biodiversity, Volume 13, Issue -, 2016, Pages 1369-1379

Abstract

The genus Centaurea L. is one of the largest and important genera of Asteraceae family. Centaurea species have been widely used as herbal remedies in folk medicine for their antidandruff, antidiarrheic, antirheumatic, anti-inflammatory, choleretic, diuretic, digestive, stomachic, astringent, antipyretic, cytotoxic, and antibacterial properties. Centaurea baseri Kose & Alan is a recently described local endemic species in Turkey and this is the first study on the chemical composition and bioactivity of its hydrodistilled essential oil and the crude extract. According to chromatospectral analysis, hexadecanoic acid (42.3%), nonacosane (8.2%), and heptacosane (8.0%) were the main compounds of the essential oil, while 16 compounds were determined in the MeOH extract using LC/MS. Furthermore, antimicrobial, antioxidant, and cytotoxic effects of the essential oil and the extract were evaluated in comparison with the standard agents. The extract showed strong antifungal effect against Candida utilis at the concentration of 60 µg/ml (MIC) where the EO showed growth inhibition at the concentration of 47.00 µg/ml (MIC) against pathogen Bacillus cereus. Both the essential oil and the extract did not show any selective antioxidant properties. The extract showed remarkably selective cytotoxic properties against MCF-7, PANC-1, A549, and C6 glioma cells.

Insecticidal activity of edible Crithmum maritimum L. essential oil against Coleopteran and Lepidopteran insects

Original Article Industrial Crops and Products, Volume 89, Issue -, 2016, Pages 383-389

Abstract

Crithmum maritimum L. (Apiaceae) is an edible plant that is used in salads or consumed as pickles in Cyprus. In our insecticidal screening study of the plant species of Cyprus, we have studied the insecticidal activity (contact and fumigant toxicity) of the essential oil of Crithmum maritimum against stored product pests Sitophilus granarius L., S. oryzae L., Tribolium castaneum Herbst., T. confusum Jacquelin du Val., Rhyzopertha dominica Fabricius., Oryzaephilus surinamensis L. (Coleoptera) and the field pest Spodoptera exigua (Hübner) (Lepidoptera). Dried leaves of C. maritimum afforded an essential oil with 0.22% (v/w) yield. The essential oil was analyzed with GC, GC–MS and the major components of the oil were identified as ?-terpinene (39.3%), ß-phellandrene (22.6%), carvacrol methylether (10.5%) and (Z)-ß-ocimene (8.2%). Highest contact toxicity of the oil was observed against S. oryzae, R. dominica and O. surinamensis (1 µL/insect application of 10% (v/v) oil solution in acetone, after 72 h, 93.30%, 83.26% and 70.33%, respectively). Highest fumigant toxicity was observed for S. granarius, S. oryzae and O. surinamensis (10 µL/10 mL container application of 10% (v/v) oil solution in acetone, after 48 h, 100.00%, 100.00% and 90.75%, respectively). Essential oil was tested on S. exigua larvae at different development stages (3rd, 4th and 5th instar larvae). 100 µL/mL essential oil was used with 1, 2 and 4 µL/larvae concentrations for 3rd, 4th and 5th instar S. exigua larvae, respectively. The toxicity of the oil against the larvae was evaluated after 24 h. C. maritimum essential oil afforded 89% mortality against the 3rd instar larvae however the mortality was decreased to 50% in the 4th and none in the 5th instar S. exigua larvae. The oil also afforded considerable AChE and BChE enzyme inhibition (50.3% and 59.8%, respectively) using the Ellman spectrophotometric method. The essential oil did not produced considerable activity (104 fold lower than the positive controls) against the selected pathogens that could be found on wheat (Bacillus cereus, B. subtilis, Salmonella typhimurium, Staphylococcus aureus, Escherichia coli) using a microdilution method for antimicrobial evaluation.

Chemical characterization and anticholinesterase effects of essential oils derived from Salvia species

Original Article Journal of Essential Oil Research, Volume 28, Issue 4, 2016, Pages 322-331

Abstract

Inhibitory effect of Salvia species herbal preparations on acetylcholinesterase (AChE) and butylcholinesterase (BuChE) activitiy may contribute to regulation of cognitive performance and impaired cholinergic functions in patients with Alzheimer’s disease. This functional role of Salvia species and their components makes the investigations on Salvia valuable in medicine-related plant research. Within this work it was aimed to investigate the in vitro anti-cholinesterase effect of essential oils derived from ten Salvia species, which grow in Turkey. The chemical composition of essential oils were characterized by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS), respectively. Results showed that all of the essential oils exhibited AChE inhibitory activity. S. pseudeuphratica, S. hydrangea and S. divaricata essential oils demonstrated the most potent AChE inhibitory effect [50% inhibition concentration (IC50) = 26.00 ± 2.00 µg/mL, 40.0 ± 4.00, 64.68 ± 4.16, respectively]. The essential oil of S. pseudeuphratica demonstrated the highest inhibitory activity against AChE and BuChE among the tested Salvia essential oils. Evidences from the our study augment the importance of essential oils obtained from Salvia species and may support utilization of Salvia species for symptomatic treatment of Alzheimer disease

The Identification of suberosin from Prangos pabularia Essential Oil and Its Mosquito Activity against Aedes aegypti

Original Article Rec. Nat. Prod., Volume 10, Issue 3, 2016, Pages 311-315

Abstract

detailed analysis of Prangos pabularia Lindl. (Apiaceae) fruit oil was performed by gas chromatography (GC-FID) and gas chromatography-mass spectrometry (GC-MS). Bicyclogermacrene (21%), (Z)-?-ocimene (19%), ?-humulene (8%), ?-pinene (8%) and spathulenol (6%) were the main constituents of the oil. One compound with 1.8% at RI 3420 remained unidentified or tentatively identified as suberosin from the Wiley GC-MS Library. The assumed compound, suberosin was synthesized in two steps and its structure was confirmed by 1D NMR and GC-MS analyses. As part of our continued research to discover new chemicals for use in mosquito control agents as repellents and larvicides, suberosin and its parent compound coumarin were investigated for the mosquito biting deterrent and larvicidal activity against Aedes aegypti. Both suberosin and coumarin showed biting deterrent activity but the activity was lower than the positive control, DEET (N,N-diethyl-3-methylbenzamide). In larval bioassays, suberosin with LC50 value of 8.1 ppm was significantly more toxic than coumarin (LC50 = 49.6 ppm) at 24-h post treatment. These results indicate that suberosin may be useful for use as mosquito larvicidal agent.

Chemical composition, antioxidant and antimicrobial activities of essential oils from leaves, aerial stems, basal stems, and rhizomes of Etlingera fimbriobracteata (K.Schum.) RMSm.

Original Article Industrial Crops and Products, Volume 84, Issue -, 2016, Pages 189-198

Abstract

Essential oils from the leaves, aerial stems, basal stems, and rhizomes of Etlingera fimbriobracteata (K.Schum.) R.M.Sm. were analyzed using a gas chromatography-flame ionization detector (GC-FID) and by gas chromatography-mass spectrometry (GC–MS). -Pinene (67.8%) and 1,8-cineole (37.2%) were the main components of the oils from leaves and aerial stems, respectively, whereas decanal was the main component of basal stems and rhizomes (27.5% and 34.4%, respectively). The antioxidant activities of the oils were evaluated using several in vitro assays; the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2 -azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging assays, the reducing power ability (RPA) assay, and the ferric reducing antioxidant power (FRAP) assay. In all the tests, essential oils from different plant parts showed very poor antioxidant activity compared with those of the reference standards, ascorbic acid (AA) and butylated hydroxytoluene (BHT). However, all the oils exhibited moderate to potent broad-spectrum antimicrobial activity against all tested strains except for Pseudomonas aeruginosa. In disc diffusion and broth microdilution assays, the inhibition zones and MIC values of the essential oils were in the range of 8–87.7 mm and 2.4–625 g/mL, respectively. Because of their strong antimicrobial activity, the essential oils of E. fimbriobracteata could be a potential alternative to conventional antimicrobials in food, cosmetics, and pharmaceutical industries.

Modulation of Human Neutrophil Responses by the Essential Oils from Ferula akitschkensis and their Constituents

Original Article Journal of Agriculture and Food Chemistry, Volume 64, Issue 38, 2016, Pages 7156-7170

Abstract

Essential oils were obtained by hydrodistillation of the umbels+seeds and stems of Ferula akitschkensis (FAEOu/s and FAEOstm, respectively) and analyzed by gas chromatography and gas chromatography-mass spectrometry. Fifty-two compounds were identified in FAEOu/s; the primary components were sabinene, ?-pinene, ß-pinene, terpinen-4-ol, eremophilene, and 2-himachalen-7-ol, whereas the primary components of FAEOstm were myristicin and geranylacetone. FAEOu/s, ß-pinene, sabinene, ?-terpinene, geranylacetone, isobornyl acetate, and (E)-2-nonenal stimulated [Ca(2+)]i mobilization in human neutrophils, with the most potent being geranylacetone (EC50 = 7.6 ± 1.9 µM) and isobornyl acetate 6.4 ± 1.7 (EC50 = 7.6 ± 1.9 µM). In addition, treatment of neutrophils with ß-pinene, sabinene, ?-terpinene, geranylacetone, and isobornyl acetate desensitized the cells to N-formyl-Met-Leu-Phe (fMLF)- and interleukin-8 (IL-8)-induced [Ca(2+)]i flux and inhibited fMLF-induced chemotaxis. The effects of ß-pinene, sabinene, ?-terpinene, geranylacetone, and isobornyl acetate on neutrophil [Ca(2+)]i flux were inhibited by transient receptor potential (TRP) channel blockers. Furthermore, the most potent compound, geranylacetone, activated Ca(2+) influx in TRPV1-transfected HEK293 cells. In contrast, myristicin inhibited neutrophil [Ca(2+)]i flux stimulated by fMLF and IL-8 and inhibited capsaicin-induced Ca(2+) influx in TRPV1-transfected HEK293 cells. These findings, as well as pharmacophore modeling of TRP agonists, suggest that geranylacetone is a TRPV1 agonist, whereas myristicin is a TRPV1 antagonist. Thus, at least part of the medicinal properties of Ferula essential oils may be due to modulatory effects on TRP channels

Original Article Journal of Oleo Science, Volume 65, Issue 10, 2016, Pages 867-870

Abstract

There is only a couple of reports indicating essential oil composition of Asphodelus species in the literature. However, from the members of this genus many non-volatile secondary metabolites were isolated. In Cyprus, Asphodelus aestivus Brot. can be found abundantly in all regions of the island. This plant has various ethnobotanical uses in Cyprus. There is no report on the volatiles nor the essential oil composition of A. aestivus. The smell of A. aestivus flowers resembles that of a cat pee which caught our attention. Therefore, we have carried out GC, GC/MS analysis of the essential oil (yield: 0.01 v/w) obtained from Asphodelus aestivus flowers. Seventeen compounds were identified in the essential oil comprising 96.2% of the oil. The major components of the essential oil were hexadecanoic acid 35.6%, pentacosane 17.4%, tricosane 13.4% and heptacosane 8.4%. In our results, we expected to see sulfur containing cat pee odorants due to the odor of the flower whereas high amounts of n-alkanes, saturated fatty acids and minor amounts of acyclic diterpenes were observed

GC-MS analysis: In vivo hepatoprotective and antioxidant activities of the essential oil of Achillea biebersteinii Afan. growing in Saudi Arabia

Original Article Evidence-based Complementary and Alternative Medicine, Volume 2016, Issue -, 2016, Pages -

Abstract

Liver disease is a worldwide problem. It represents one of the main causes of morbidity and mortality in humans. Achillea biebersteinii is used as herbal remedy for various ailments including liver diseases. But the scientific basis for its medicinal use remains unknown. Thus, this research was undertaken to evaluate the efficiency of A. biebersteinii essential oil (ABEO) (0.2?mL/kg) in the amelioration of CCl4-induced hepatotoxicity in rodent model. Moreover, the chemical content of the oil was investigated using GC and GC-MS. The following biochemical parameters were evaluated: serum glutamic oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), gamma-glutamyl-transpeptidase (?-GGT), alkaline phosphatase (ALP), and total bilirubin. Furthermore, lipid profile, malondialdehyde (MDA), nonprotein sulfhydryl (NP-SH), and total protein (TP) contents in liver tissue were estimated. 44 components (92.0%) of the total oil have been identified by GC-MS analysis where ?-terpinene and -cymene were the most abundant. The high serum enzymatic (GOT, GPT, GGT, and ALP) and bilirubin concentrations as well as the level of MDA, NP-SH, and TP contents in liver tissues were significantly reinstated towards normalization by the ABEO. Histopathological study further confirmed these findings. In addition, ABEO showed mild antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ß-carotene-linoleic acid assays.
