Abstract

In this experiment, effectiveness of antibiotic treatment was evaluated by bacteriology and Somatic Cell Count (SCC). Ninety-six heifers were divided randomly into two groups: either to be treated with antibiotics (200 mg cephalexin monohydrate and 250 mg neomycin) 45 days prior to expected calving date (Group 1, n = 41) or not treated (Group 2, n = 55). Bacteriologic tests were used for detection of infected quarters. Of the quarters, 58.3% had infection at least in one quarter before calving. Mastitis pathogens were isolated from 31% of the quarters. In the treatment group, 43 of 52 infected quarters (82.6%) were cured but new infections (n = 43) were detected in the beginning of the lactation. Spontaneous cure rate was 69.3% in the control group and new infection rate (24.2%) was lower than the treated group. The treatment was effective for reducing prepartum Intramammary Infections (IMI) and persistent IMI; it was however inadequate to protect the quarters from new IMI in the beginning of lactation (p>0.05). Precalving antibiotic treatment was however quite effective for reducing individual SCC. There were less quarters (n = 5) with high SCC (>400.000 cells mL-1) in the treated group than the control group. In conclusion, antibiotic treatment prepartum may be necessary to reduce incidence of mastitis and improve quality of milk.

Staphylococci and threshold value of somatic cell count for diagnosis of sub-clinical mastitis in Pirlak Sheep at mid-lactation

Abstract

This study investigated the bacterial agents causing sub-clinical mastitis and the mean somatic cell counts (SCC) of milk in Pirlak sheep at mid-lactation. The percentage of infected udder halves was 11.4% (53 / 464). The most frequently isolated species were coagulase-negative staphylococci (CNS) (64.2%), followed by Staphylococcus aureus (24.5%) and Escherichia coli (11.3%). Among the CNS, the most common species was Staphylococcus epidermidis (38.2%). The other species isolated from milk samples were Staphylococcus xylosus (17.7%), Staphylococcus chromogenes (14.7%), Staphylococcus simulans (8.8%) and Staphylococcus hyicus (8.8%). The mean SCC for culture positive and negative samples was 1742 · 103 and 161 · 103 cells / ml, respectively. A significant difference (p < 0.05) was determined between with and without microbial growth groups in terms of the SCC values. Threshold limit for SCC was 374 · 103 cells / ml for Pirlak sheep. In conclusion, it was considered that SCC is an important predictor of sub-clinical mastitis in Pirlak sheep. This is the first study to describe the bacterial agents causing sub-clinical mastitis and threshold limit for SCC in Pirlak sheep in Turkey.

Abstract

This study was carried out for detection of nitric oxide (NO) levels in healthy (Group 1, n:78) and infected (Group 2, n:63) quarters of pregnant heifers. The mean nitric oxide (NO) concentrations were calculated of about 15,58 ± 2,21 µM/ml and 42,69 ± 6,03 µM/ml for Group 1 and Group 2, respectively. A significant difference was found between two groups, statistically (p<0,001). The mean NO concentrations were compared between quarters infected with Staphylococcus aureus (S. aureus) and coagulase negative staphylococcus (CNS) in Group 2 and measured to be; 38,50 ± 8,47 µM/ml and 44,17 ± 7,71 µM/ml, respectively (p>0,05). Additionally, lacteal secretions were classified to be thickened and watery according to viscosity. Intramammarial infection rates (IMI) and NO concentrations were compared in low (watery) and high (thickened) viscosity samples. Risk of IMI was found higher in thickened samples than watery (p<0,001). Every 1 unit (µM/ml) increase in the NO concentration boosted the risk of IMI 1,036-fold (p<0,001).

Bu çalışma gebe düvelerin sağlıklı (Grup 1, n:78) ve enfekte (Grup 2, n:63) meme loblarında nitrik oksit düzeylerini belirlemek için planlandı. Grup 1 ve Grup 2 deki ortalama nitrik oksit düzeyleri sırasıyla 15,58 ± 2,21 µM/ml ve 42,69 ± 6,03 µM/ml olarak belirlendi ve iki grup arasında NO düzeyi bakımından istatistiki olarak belirgin bir fark bulundu (p<0,001). Grup 2’de Staphylococcus aureus ve koagulaz negatif stafilokoklar ile enfekte meme loblarındaki ortalama NO düzeyleri kıyaslandı ve sırasıyla; 38,50 ± 8,47 µM/ml ve 44,17 ± 7,71 µM/ml olarak ölçüldü (p>0,05). Nitrik oksit düzeyine ek olarak lakteal sekresyonları viskozitesine göre yoğun ve sulu olarak sınıflandırıldı. Düşük (sulu) ve yüksek (yoğun) viskoziteli örneklerde meme içi enfeksiyon oranları ve NO konsantrasyonları kıyaslandı. Yoğun örneklerde meme içi enfeksiyon riski, sulu örneklerden daha yüksek bulundu (p<0,001). Nitrik oksit konsantrasyonundaki bir birimlik artışın, enfeksiyon riskini 1,036 kat artırdığı belirlendi (p<0,001).

Abstract

We investigated eight adult dogs that were brought to veterinary clinics with a history of transmissible venereal tumors (TVT). Our goal was to demonstrate the occurrence of apoptosis and the cessation of cell proliferation at every phase of scheduled chemotherapy for naturally occurring TVT. Tissue samples were collected immediately after weekly treatments with vincristine sulfate and processed for histological purposes. Sections 5 µ m thick were stained by the TUNEL reaction for apoptosis and immunostained for Ki67 as a proliferation marker. We observed that after vincristine applications, tumor cell proliferation ceased and apoptosis increased. Ki67 HSCORE values were signifi cantly lowered after the fi rst and second treatments with the chemotherapeutic agent compared to controls, whereas TUNEL HSCORE values were signifi cantly higher after two applications of vincristine compared to controls. Our results suggest that scheduled vincristine sulfate applications stabilize the induction of tumor regression by inducing apoptosis and preventing cell proliferation.

Abstract

The aim of this study was to evaluate nitric oxide (NOx) concentration in infected and non-infected mammary quarters of dairy heifers before and after calving. The relationship between bacterial species and NOx concentrations, as well as correlation between NOx concentrations and postpartum somatic cell count (SCC), was assessed. Coagulase-negative staphylococci, Staphylococcus aureus and Escherichia coli were the bacteria commonly isolated during the pre- and postpartum period. Infected quarters had greater NOx concentrations than non-infected quarters before (30.81 v. 22.83 mM/ml, P,0.05) and after (9.56 v. 5.77 mM/ml, P,0.0001) calving. It was determined that the interaction between sampling period and infectious status had no effect on NOx concentration ( P,0.16). Infected quarters had greater SCC (log10) than healthy quarters (4.95 v. 4.39; P,0.0001). NOx concentrations, however, did not correlate with SCC (r50.02). In summary, changes in NOx concentration were mainly dependent on the infectious status of the quarters with variations among the bacterial species ( P,0.05).

Abstract

The aim of this study was to determine the effectiveness of dry-off antibiotic therapy and teat sealant on somatic cell count (SCC) in Saanen goats. The goats were randomly divided into 3 groups. In Groups I (n = 50) and II (n = 50), the goats were treated with intramammary antibiotics and a combination of intramammary antibiotics and internal teat sealant at dry-off, respectively. The animals in Group III (n = 50) were designated as the control group. For the SCC analysis and bacteriological examination, milk samples (n = 900) were collected separately from each udder half during routine morning milking, prior to drying off, and at months 1, 2, 3, 4, and 5 of following lactation. The pretreatment prevalence of intramammary infection at dry-off was 4.7% (7/150) for halves. Isolated pathogens were coagulase-negative staphylococci (57.1%) and S. aureus (42.9%) from infected halves. Dry-off antibiotic and dry-off antibiotic with teat sealant therapy reduced milk SCC levels significantly (P < 0.01) when compared to the control group. On the other hand, the difference in SCC between dry-off antibiotic and dry-off antibiotic with teat sealant therapy groups was not statistically significant (P > 0.01). In conclusion, intramammary antibiotics administered alone and a combination of intramammary antibiotics and teat sealant reduced milk SCC levels significantly

Corpus Luteum Insufficiency in Dogs (Canis lupus)

Abstract

Insufficient progesterone synthesis, so called hypoluteoidism or luteal insufficiency, is one of the possible reasons for infertility in the bitch. The bitch ovulates at progesterone concentrations of about 5-10 ng/ml. The concentrations increase to >25 ng/ml within 3-4 weeks and then subsequently decrease. In the pregnant bitch, progesterone rapidly drops to <2 ng/ml approximately 24-48 hours before parturition. Luteal insufficiency, characterized as an early decrease of progesterone secretion, is most commonly observed between days 20 and 35 of pregnancy.Lower concentrations result in resorption and abortion, respectively. In bitches suspected to have luteal insufficiency, weekly progesterone determinations using quantitative tests should be performed from 5-7 days after mating or at least from the date of early pregnancy diagnosis. The frequency has to be increased in the case of progesterone concentrations below 10 ng/ml. Either natural or synthetic progestins can be used. Administration of natural progesterone should be stopped 2-3 days before expected parturition otherwise it would result in a prolonged duration of pregnancy with dystocia and stillbirth.

Progsteronun yetersiz sentezi (hipoluteoidizm, luteal yetersizlik) köpeklerde infertiliteye neden olurken, tanısında östrüs siklusundaki progesteron konsantrasyonu değişikliklerini dikkate almak gerekmektedir. Dişi köpeklerde ovulasyon 5-10 ng/ml serum konsantrasyonunda şekillenirken bunu takip eden 3-4 hafta içerisinde değerler 25 ng/ml?nin üzerine kadar yükselir ve sonrasında düşmeye başlar. Gebe köpeklerde antepartum 24-48 saatlerinde progesteron konsantrasyonu gebeliğin devamı için kritik sınır olan 2 ng/ml?in altına düşerken corpus luteum?dan salınan progesteron sekresyonunun erken dönemde azalması luteal yetmezliğe neden olmaktadır. Luteal yetmezlik çoğunlukla gebeliğin 20 ile 35. günleri arasında ortaya çıkarken, rezorpsiyon ve aborta neden olabilmektedir. Luteal yetmezlik şüphesi bulunan dişi köpeklerde, çiftleşmeden yaklaşık 5-7 gün sonra başlayarak haftalık progesteron ölçümü yapılmalıdır. 10 ng/ml?nin altındaki değerlerde muayene aralığı artırılmalıdır. Canlı fetüs tespit edilen gebelik süresince progesteron değerinin istenilenin altına düşmesi durumunda doğal veya yapay progestin desteği gerekmektedir. Doğal progesteron uygulaması, hesaplanan doğum tarihinden 2-3 gün önce durdurulmalıdır, aksi takdirde uzayan gebelik süresine bağlı olarak gebelik süreci güç veya ölü doğum ile sonuçlanabilir.
