    EXPERIMENTAL INVESTIGATION OF RATS DEVELOPED PROTECTIVE EFFECT OF MELATONIN IN THE ISCHEMIC HEART FAILURE

    In this study; we reported preventive effect of melatonin on heart failure as a result this agent which isn’t toxic clinically is supposed to generate new principles for treatment.

    It is well known during myocardial infarction and subsequently reactive oxygen species can be produced and increasing of these species promotes the current necrosis to lead heart failure. The aim of this study was to research probable protective effect of melatonin on the ischemic heart failure that generated through the ligation of left anterior descending coronary artery (LAD). Wistar albino rats were anaesthetized by ketamine and left anterior descending coronary artery of rats were occluded to generate ischemic heart failure and then animals observed for 4 weeks. Through the experiment; rats were treated with normal saline and melatonin (Mel:10 mg/kg), 24 hours after the last administration rats were assessed by echocardiography, their blood presures and heart rates were evaluated. In addition, their body weight were measured before and after treatment. At the end of trial, animals were decapitated to take blood and tissue sample. Heart were weighted when they were wet and after dried than heart weight (HW:mg) was determined. HW/BW rate was estimated as hypertrophy index. Lactate dehydrogenase(LDH), creatine kinase (CK), lysosomal enzymes included ß-D glucuronidase, ß-D galactosidase ß-D-N acetyl glucose aminidase acid phospotase ve cathepsin D were determinated at the blood sample. In addition to this, Na-K ATPase, ATP, malondialdehyde (MDA), glutathione (GSH), myeloperoxidase (MPO) levels were also determinated at heart. Caveolin-3 and sarcoplasmic/ endoplasmic reticulum Ca2+ ATPase (SERCA) levels were evaluated by histological techniques and western blot analyse. According to our blood analyses; ischemic heart failure significantly increased the activities of CK, LDH and lysosomal enzymes. Due to ischemic heart failure, levels of MDA, and activities of caspase-3 and MPO were found significantly higher than the control groups, on the other hand GSH, ATP, caveolin-3, SERCA and Na-K ATPase activity were found low. Nevertheless, findings of echocardiography and histology indicated structural and functional damage. All paremeters were refined with treatment of melatonin and histological recovery were determined. In this study; we reported preventive effect of melatonin on heart failure as a result this agent which isn’t toxic clinically is supposed to generate new principles for treatment.

    Effects of melatonin and 5-methoxytryptophol on synovial inflammation in the zymosan-induced rheumatoid arthritis in rats

    MEL and 5-MTX reducing inflammation in arthritis suggests these agents might constitute a new therapeutic principle clinically.

    The aim of this study is the investigation of synoviocytes, cytokines and metalloproteinases in the synovial inflammation model which is created by zymosan application to temporomandibular joint (TMJ) and effects of pineal hormones melatonin (MEL) and 5-methoxytryptophol (5-MTX) on these parameters. 200-250 g Wistar albino rats of both sexes were used for modeling arthritis in this study. Arthritis model was created by intraarticularly (i.a.) injecting 2 mg zymosan in 40 ml saline into the left temporamandibular joint of the rats while the sham group was created by only injecting 40 ml saline solution (intraarticulary). MEL and 5-MTX administration was made intraperitoneal before zymosan injection. 6 hours after zymosan or saline administration of MEL and 5-MTX the synovial fluid was collected from the animals and the synovial membrane was collected for histological assessment. The administration of zymosan increased the release of IL-1ß and TNF? and the activity of metalloproteinases (MMM-9) and metalloproteinases-2 (MMP-2) and with this administration values got closer to the sham group. The histological evaluation showed significant increase in the intensity of synoviocytes that arose in the inflammation was found to subside. In conclusion, MEL and 5-MTX reducing inflammation in arthritis suggests these agents might constitute a new therapeutic principle clinically.

    INVESTIGATION OF RATS DEVELOPED DUE TO BURN INJURY AGAINST LUNG PROTECTIVE EFFECT BETAINE

    In this study; we reported preventive effect of betaine on lung injury that be constitued with burn; as a result this agent which isn’t toxic clinically is supposed to generate new principles for treatment.

    Mechanisms of burn-induced lung injury involve oxidant generation and the release of pro-inflammatory cytokines. Animal models of burn injury indicate oxygen radicals as causative agents in local wound response, as well as in the development of burn shock. This study was designed to determine the possible protective effect of betaine treatment against oxidative damage in the lung induced by burn injury. Under ether anaesthesia, the shaved dorsum of Wistar albino rats was exposed to a 90°C water bath for 10s to induce burn injury. Betaine was administered p.o (250mg / kg) for a period of 21 days before burn injury and single dose of betaine was administered after burn injury. In the sham group the same protocol was applied in the expect that the dorsum was dipped in a 25°C water bath for 10s. At the end of experiment rats were decapitated and blood was collected for the analysis of pro-inflammatory cytokines [tumor necrosis factor-? (TNF- ?) and interleukin 1 beta (İL-1ß)], lactate dehydrogenase (LDH) activity. Lung tissue samples were taken for the determination of malondialdehyde (MDA) and glutathione (GSH) levels, myeloperoxidase (MPO) and Na+, K+-ATPase activity in addition to the histological analysis. Burn injury caused significant increase in both cytokine levels and LDH activity. On the other hand, in lung tissue the raised MDA levels, MPO activity and reduced GSH levels, Na+-K+ ATPase activity due to burn injury were found.Treatment of rats with betaine significantly elevated the reduced GSH level and Na+,K+-ATPase activity while it decreased MDA level and MPO activity. All of changes in serum and tissue caused by burn showed a significant improvement with administration of betaine. In this study; we reported preventive effect of betaine on lung injury that be constitued with burn; as a result this agent which isn’t toxic clinically is supposed to generate new principles for treatment.

    Investigated the protective effects of resveratrol on myocardial infarction induced by isoproterenol in rats

    In this study; we reported preventive effect of resveratrol on myocardial ınfarction that be constitued with ısoproterenol; as a result this agent which isn’t toxic clinically is supposed to generate new principles for treatment.

    Ischemia reperfusion injury that occur as an effect of sympathetic hyperactivity , known to play an important role in myocardial infarction. In this study we researched preventive effect of resveratrol on myocardial infarction that be constituted with ısoproterenol (ISO) which is an synthetic non-selective ß-reseptor agonist. Rats were treated with intraperitonal 5mg/kg Isoproterenol for 30 days to constitute myocardial infarction and concurrently they were treated with normal saline and resveratrol. At the end of trial, after 24 hours of the last administration, rats were anaesthetized with pentobarbital (35mg/kg ı.p) and echocardiographic parameters included left ventricle wall thickness,diastolic dimension anterior wall thickness, fractional shortening diastol dimension were evaluated with echocardiography and after 48 hours of the last administration blood pressure and heart rate evaluated. In addition, their body weight were measured before and after treatment. At the end of trial, animals were decapitated to take blood and tissue sample. Hearts were meausured after they were wet and dried and heart weight (HW:mg)was determined. HW/BW rate was estimated as hypertrophy index. Lactate dehydrogenase(LDH), creatine kinase (CPK), lysosomal enzymes included ß-D glucuronidase, ß-D galactosidase ß-D-N acetyl glucose aminidase acid phospotase ve cathepsin D were determinated at the blood sample. In addition to this, Na-K ATPase ,ATP ,MDA,GSH,MPO and kollagen determinated at heart and caveolin-3 levels were evaluated with histological techniques and western blot analyse. According to our blood analyses ISO significantly increased the activities of CK, LDH and lysosomal enzymes. Due to ISO, levels of MDA, and activities of caspase-3 and MPO were found significantly higher than the control groups, in contrast levels of GSH, ATP, caveolin-3 and activity of Na-K ATPase were found low. Nevertheless, findings of echocardiography and histology indicated structural and functional damage. Abstract

Background Curcumin has been shown to decrease ischemia-reperfusion (I/R) injury in kidney or brain tissues. In this study, the effects of curcumin were evaluated in skeletal muscle during I/R injury. Materials and Methods Hind limb ischemia was induced by clamping the common femoral artery and vein. After 4 h ischemia, the clamp of the femoral vessels of animals was taken off and the animal underwent 2 h reperfusion. We measured plasma concentrations of interleukin-1ß (IL-1ß) and tumor necrosis factor-? (TNF-?) using enzyme-linked immunosorbent assay (ELISA). The right gastrocnemius muscle was harvested and immediately stored at -30°C for the assessment of superoxide dismutase (SOD), catalase (CAT) activities, and measurement of glutathione (GSH), malondialdehyde (MDA), and protein oxidation (PO) levels. Curcumin (100 mg/kg), ?-tocopherol, and normal saline (10 mL /kg1) were administered intraperitoneally 1 h prior reperfusion. Results Plasma TNF-? or IL-1ß levels increased significantly in I/R group. The plasma levels of these proinflammatory cytokines were reduced in curcumin group. Muscle tissues of I/R groups revealed significantly higher antioxidant enzyme (superoxide dismutase, glutathione peroxidase, catalase) activities, and increased levels of malondialdehyde, nitric oxide, and protein carbonyl content compared with the SHAM group. Levels of these parameters in muscle revealed significant reductions in the I/R + curcumin group compared witho the I/R group. Curcumin has more potent antioxidant activity than vitamin E in the skeletal muscle I/R. Conclusion In this study, protective effects of curcumin against skeletal muscle ischemia-reperfusion injury have been revealed. We underscore the necessity of human studies with curcumin that would be hypothetically beneficial preventing skeletal muscle I/R injury.

Efficacy of pentoxifylline and tadalafil in rat model of ischemic colitis.

Abstract

Objective: The aim of this study is to investigate the efficacy of tadalafil against pentoxifylline in rat model of ischemic colitis (IC). Material-Methods: Thirty-two Wistar albino rats were subjected to laparotomy and left colon devascularization to create an IC model and then randomly placed into four groups. Group-1 (sham group) was administered 0.9% NaCl following laparotomy, group 2 (control group) was administered 0.9% NaCl following induced IC, group 3 was given pentoxifylline (n = 8), and group 4 was given tadalafil. On the third day; macroscopic findings, Gomella's ischemic area and Wallace scoring, histopathological analysis, and Chiu scoring were performed, and malondialdehyde (MDA) measurement in ischemic colon tissue was carried out through chemical analysis. Results: Significant differences were observed in acidic fluid, bowel dilatation, and serosal change (p < .05). The ischemic area measured 63.3 mm2 in the control group, 2.8 mm2 in the pentoxifylline group (p = .0001), and 2.4 mm2 (p = .0001) in the tadalafil group. A significant difference was seen between the sham group and the control and pentoxifylline groups (p < .01), in terms of Wallace score and Chiu classification. Similarly, a significant difference was determined between the control group and pentoxifylline and tadalafil groups (p < .01), but no significant difference was established between the pentoxifylline group and tadalafil group (p = .33). MDA measurement was found on an average to be 63.7 in the control group, 22.7 in group 3 and 22.8 in group 4 (p = 001). Conclusion: Although tadalafil is superior to pentoxifylline, both drugs are considered to have positive effects.

Effect of hyperbaric oxygen treatment in a rat model of abdominal aortic aneurysm.

Abstract

Matrix metalloproteinase (MMP) plays a pivotal role in the pathophysiology of abdominal aortic aneurysms (AAA). Experimental studies have demonstrated that hyperbaric oxygen (HBO2) therapy decreases MMP levels in different tissues. However, the effect of HBO2 therapy on AAA has yet to be investigated. This study aimed to examine the effects of HBO2 on MMPs in an experimental AAA model. The model was implemented with CaCl2 in 12-week-old male Wistar albino rats. The rats were randomized into four groups: Group I: received NaCl (n = 6) (Sham group); Group II: received NaCl and were treated with HBO2 (n = 6); Group III: received CaCl2 (n = 6); and Group IV: received CaCl2 and were treated with HBO2 (n = 6). HBO2 therapy was applied for five of seven days over a period of six weeks. Although in the CaCl2 groups, aortic diameters were significantly higher than the NaCl groups (p < 0.05), there was no difference between pre- and post-HBO2 in the CaCl2 groups (p > 0.05). In the CaCl2 group, the MMP-2 and MMP-9 levels were significantly higher than those in the NaCl group (p < 0.05). HBO2 therapy had no statistically significant effect on the MMP-2 and MMP-9 levels in Groups III and IV. However, it was observed that both levels clearly decreased in Group IV. In conclusion, the study suggested that HBO2 may have favorable effects on MMP levels.

Effects of inactivated Parapoxvirus ovis on polymorphonuclear leukocyte function and myeloperoxidase activity in horses

Abstract

Immunomodulatory products have been used for years in veterinary medicine. Inactivated Parapoxvirus ovis (iPPVO) is currently used in equine medicine as an immunomodulator to improve the immune system and as a prophylactic treatment to prevent or treat infectious diseases. This study was designed to determine the effects of iPPVO on polymorphonuclear leukocyte (PMNL) function (phagocytosis and intracellular killing activity) and the myeloperoxidase (MPO) activity of PMNLs in horses. Twenty-four healthy English thoroughbred horses with an average age of 11 years were included in the study. Venous blood samples (10 ml) were taken before (agent-free controls) and after the administration of iPPVO (2 ml i.m. injection on Days 1, 3, and 5). PMNLs (1 x 107 cells/ml) were isolated from venous blood containing EDTA (0.1 g/ml) with Ficoll-Hypaque gradient centrifugation. Cellular phagocytosis and intracellular killing activities were assayed using a modification of Alexander's method before and after treatment with iPPVO. MPO activity was also measured. The administration of iPPVO significantly increased the phagocytic, intracellular killing, and MPO activities of equine PMNLs (P = 0.0058, P = 0.0050, and P = 0.0070, respectively). This study demonstrates a strong correlation between MPO activity and PMNL function. The administration of iPPVO to horses has a supportive effect on their cellular immunity and an immunomodulatory effect against equine viral infections.

Objective: Nephrotic syndrome (NS) may result in renal failure. Apelin (AP), a vasoactive peptide, demonstrates its effect by binding to the AP receptor. AP, present in the endothelial and vascular smooth muscle cells of glomerular arterioles, affects the pre- and post-microvascularization by regulating renal hemodynamics. This study aimed to determine the histomorphological changes of AP treatment in a doxorubicin (DOX)-induced NS model. Methods: Male Sprague Dawley rats were used. Rats were randomly divided into four groups (n=6): control [physiological saline (PS) solution intraperitoneal (i.p.)]; AP+PS (PS and 50 mcg/kg/day AP-13 i.p.); NS [10 mg/ kg DOX i.p.] and NS+AP (NSAP; 10 mg/kg DOX+50 mcg/kg/day AP-13 i.p.). Renal tissues were collected on the 22nd day for light microscopic investigations. Results: Light microscopic investigations showed that the NS group revealed adhesions between the tuft and Bowman’s capsule, mesangial matrix accumulation in the glomeruli, and tubular damage with dilatation and cast accumulation. In the NSAP group, minimal regression in the glomerular and tubular damage was observed. Conclusion: The histomorphological changes of AP treatment in a DOX-induced NS model demonstrated a limited therapeutic effect on glomerular and tubular damage in renal tissues.

Effects of melatonin and 5-methoxytryptophol on synovial inflammation in the zymosan-induced rheumatoid arthritis in rats

Abstract

The aim of this study is the investigation of synoviocytes, cytokines and metalloproteinases in the synovial inflammation model which is created by zymosan application to temporomandibular joint (TMJ) and effects of pineal hormones melatonin (MEL) and 5-methoxytryptophol (5-MTX) on these parameters. 200-250 g Wistar albino rats of both sexes were used for modeling arthritis in this study. Arthritis model was created by intraarticularly (i.a.) injecting 2 mg zymosan in 40 ml saline into the left temporamandibular joint of the rats while the sham group was created by only injecting 40 ml saline solution (intraarticulary). MEL and 5-MTX administration was made intraperitoneal before zymosan injection. 6 hours after zymosan or saline administration of MEL and 5-MTX the synovial fluid was collected from the animals and the synovial membrane was collected for histological assessment. The administration of zymosan increased the release of IL-1ß and TNF? and the activity of metalloproteinases (MMM-9) and metalloproteinases-2 (MMP-2) and with this administration values got closer to the sham group. The histological evaluation showed significant increase in the intensity of synoviocytes that arose in the inflammation was found to subside. In conclusion, MEL and 5-MTX reducing inflammation in arthritis suggests these agents might constitute a new therapeutic principle clinically.

Effect of Green Tea Extract and Whey Proteins on Experimental Gastric Ulcer Model in Rats

Abstract

Green tea has multiple effects on human health while whey proteins exhibit antioxidantive and supporting effects on immunity. In this present study, preventive and curative effects of green tea and whey proteins were investigated on rat gastric ulcer model. To determine their effects, experiments were planned at two stages involving 10-dayadministration. Lipid peroxidation, glutathione, myeloperoxidase, reactive oxygen species, levels of TNF-? and IL-1ß and histological evaluations were determined in collected tissues. As a result, combined consumption of these foods reduced the production of reactive oxygen species and lipid peroxidation, and this type of consumption had a synergistic effect for the prevention of gastric ulcer in a rat model. Biochemical and histological data in assessed tissue damages were also compatible. In conclusion, although their curative and preventive properties for gastric ulcer were significant, combined consumption of green tea and whey proteins can be considered as “functional food alternative” to support clinical therapy; however, they should not replace drug therapies.

Abstract

BACKGROUND: This study was designed to determine possible protective effect of betaine treatment against oxidative injury in pulmonary tissue induced with thermal trauma. METHODS: Under ether anesthesia, shaved dorsum of Wistar albino rats was exposed to a 90°C water bath for 10 seconds to induce burn injury. Betaine was administered orally (250 mg/kg) for a period of 21 days before burn injury, and single dose of betaine was administered after thermal injury. Control group rats were exposed to 25°C water bath for 10 seconds. Upon conclusion of experiment, rats were decapitated and blood was collected for analysis of pro-inflammatory cytokines and lactate dehydrogenase (LDH) activity. Lung tissue samples were taken to determine malondialdehyde (MDA) and glutathione (GSH) levels, myeloperoxidase (MPO), and Na+/K+-ATPase activity, in addition to histological analysis. RESULTS: Burn injury caused significant increase in both cytokine levels and LDH activity. In lung samples, raised MDA levels, MPO activity, and reduced GSH levels and Na+/K+-ATPase activity were found due to burn injury. CONCLUSION: Treatment of rats with betaine significantly restored GSH level and Na+/K+-ATPase activity, and decreased MDA level and MPO activity. According to the findings of the present study, betaine significantly diminishes burn-induced damage in tissue.


Özet

Effects of ghrelin on colonic anastomosis following radiotherapy

Abstract

Background: Ghrelin has been reported to have properties such as inducing proliferation, migration and angiogenesis of vascular endothelial cells and has anti-inflammatory effects which may help wound healing in anastomosis following neoadjuvant radiotherapy. Materials and methods: Rats were randomized into 4 groups: control group (1) underwent colonic anastomosis without treatment; ghrelin group (2) which received a single dose of ghrelin following colonic anastomosis; radiation group (3) which recieved a single dose of radiation 5 days before anastomosis; radiation + ghrelin group (4) which received a single dose of radiation 5 days before anastomosis and ghrelin following anastomosis. Anastomotic bursting pressures of the rats which were sacrificed 7 days after surgery were measured. The anastomotic segment was extracted for tissue hydroxyproline, luminol and lucigenin measurements, and histopathological examination as well. Blood samples were obtained for measurement of TNF-? and IL-1ß plasma levels. Results: Bursting pressures of anastomosis and hydroxyproline levels were significantly higher in control and radiation + ghrelin groups than in radiation group. Tissue luminol and lucigenin levels as well as plasma TNF-? and IL-1ß levels were significantly lower in control and radiation + ghrelin groups than in radiation group. Histopathological assessment revealed that administration of ghrelin resulted in regression of inflammation which was severe in radiation group. Conclusion: The adverse effects of radiotherapy on mechanic and histological parameters of anastomosis healing were attenuated by ghrelin therapy by reducing plasma proinflammatory cytokines and tissue reactive oxygen radicals.

Abstract

Objective: Agmatine, the decarboxylation product of arginine produced by arginine decarboxylase, is a novel neurotransmitter and exists in the mammalian brain. Agmatine has been reported to modulate cognitive functions, including learning and memory. Methods: In the present study, we evaluated the effects of agmatine on cognitive performance and oxidative damage in intracerebroventricular (i.c.v.) streptozotocin (STZ) model of Alzheimer’s disease (AD). Adult male Sprague-Dawley rats were injected STZ (3mg/kg, i.c.v, bilaterally) on days 1 and 3. The learning and memory patterns were assessed by using passive avoidance, Morris water maze, and closed field activity tests. Also, malondialdehyde (MDA), glutathione (GSH) levels and myeloperoxidase (MPO) activity have been determined as the parameters of oxidative damage. The behavioral tests were performed after 14 days from the first injection of STZ. Rats with impaired learning and memory performance were treated with intraperitoneal (i.p.) agmatine (40 mg/kg) twice daily for 7 days. After agmatine treatment, rats were subjected to the aforementioned behavioral tests again. Immediately after decapitation of the rats, the brains were collected and analyzed for oxidative damage parameters. Results: Agmatine improved the STZ-induced both spatial and emotinal memory impairment and oxidative damage. Findings of the study demonstrated the effectiveness of agmatine in reversing the cognitive deficits as well as the oxidative damage caused by i.c.v STZ. Conclusion: Taken together, our results have provided experimental evidence suggesting a possible therapeutic potential of agmatine as a regulator in etiopathogenesis of neurodegenerative diseases such as Alzheimer’s disease.
